Identification of SNP Markers Associated with Grain Quality Traits in a Barley Collection (Hordeum vulgare L.) Harvested in Kazakhstan

چکیده
Barley (Hordeum vulgare L.) is a cereal crop traditionally used in animal feed, malting, and food production. In this study, collection of barley was analyzed according to key grain quality traits, including protein content (GPC), starch (GSC), extractivity (EX), test weight per liter (TWL). A genome-wide association study (GWAS) conducted identify the quantitative trait loci (QTLs) associated with GPC, GSC, EX, TWL using 658 accessions from USA Kazakhstan. The grown at three breeding organizations Kazakhstan 2010 2011 genotyped 9K SNP Illumina chip. As result, 18 marker-trait associations (MTAs) for 19 MTAs 12 27 were detected, resulting 30 identified QTLs. It shown that genetic locations 25 these QTLs similar positions genes previously reported scientific literature, suggesting 5 remaining are novel putative factors studied traits. Five most significant markers (p < 2.6 × 10−5) traits GWAS development reliable informative competitive allele-specific PCR (KASP) genotyping assays. effectiveness two assays (ipbb_hv_6 ipbb_hv_128) confirmed via validation separate lines large field plots northern Therefore, KASP can be efficiently marker-assisted selection breeding.

Identification of a Phytase Gene in Barley (Hordeum vulgare L.)
BACKGROUND Endogenous phytase plays a crucial role in phytate degradation and is thus closely related to nutrient efficiency in barley products. The understanding of genetic information of phytase in barley can provide a useful tool for breeding new barley varieties with high phytase activity. METHODOLOGY/PRINCIPAL FINDINGS Quantitative trait loci (QTL) analysis for phytase activity was condu...
متن کاملAssociation analysis for traits associated with powdery mildew tolerance in barley [Hordeum vulgare L.] using AFLP markers
Association analysis is a useful method for evaluation of significant association between molecular marker and phenotype of trait. This study was performed to evaluate association between traits related with powdery mildew resistance and molecular markers. This investigation was performed using 77 barley genotypes and AFLP markers. In phenotypic evaluation, reaction of seedlings to powdery mild...
متن کاملResistance genes in barley (Hordeum vulgare L.) and their identification with molecular markers.
Current information on barley resistance genes available from scientific papers and on-line databases is summarised. The recent literature contains information on 107 major resistance genes (R genes) against fungal pathogens (excluding powdery mildew), pathogenic viruses and aphids identified in Hordeum vulgare accessions. The highest number of resistance genes was identified against Puccinia h...
متن کاملLysine Catabolism in Barley (Hordeum vulgare L.).
Lysine catabolism in seedlings of barley (Hordeum vulgare L. var. Emir) was studied by direct injection of the following tracers into the endosperm of the seedlings: aspartic acid-3-(14)C, 2-aminoadipic acid-1-(14)C, saccharopine-(14)C, 2,6-diaminopimelic acid-1-(7)-(14)C, and lysine-1-(14)C. Labeled saccharopine was formed only after the administration of either labeled 2,6-diaminopimelic acid...
متن کاملDetection of QTLs for salt tolerance in Asian barley (Hordeum vulgare L.) by association analysis with SNP markers
Two hundred ninety-six Asian barley (Hordeum vulgare L.) accessions were assessed to detect QTLs underlying salt tolerance by association analysis using a 384 single nucleotide polymorphism (SNP) marker system. The experiment was laid out at the seedling stage in a hydroponic solution under control and 250 mM NaCl solution with three replications of four plants each. Salt tolerance was assessed...
